Care and De-flasking of Sterile Plant Tissue Culture Tubes Copyright 2002, Aaron J. Hicks
If you have received 25mm diameter x 100 mm (about 1” diameter x 4” long) orchid tissue cultures, this will help you remove them from flask (de-flasking), pot them up, and help them to survive.
As you have received them, the plants in sterile culture are sensitive to their
environment. They have been raised in very high humidity (about 100%),
and without any bacteria or fungi that can harm them. If the tubes are not jumbled, and there is no sign of contamination, the plants may be left in the tubes for a period of time. Give
them low light, and keep them at a comfortable temperature. Do not leave them in the sun, cook them, or freeze them!
If contaminated, the plants may quickly be overgrown with white, green, or other-colored mold. Remove plants from contaminated tubes quickly. You may wish to immediately de
-flask plants that have grown large or are starting to develop brown leaves.
