Modification of Malmgren’s medium for germination of European species

Sucrose 18 g Agar 6 g MgSO4 * 7 H2O 150 mg CaCl2 (anhydrous) 90 mg KH2PO4 (anhydrous) 150 mg FeSO4 * 7 H2O 18 mg myo-inositol 100 mg Peptone 4 g Pineapple juice 25 mL Harvais micronutrients 1 mL (see below)

Harvais micronutrients (1000x)

H3BO3 500 mg CuSO4 * 5 H2O 25 mg ZnSO4 * 7 H2O 500 mg Na2MoO4 * 2 H2O 20 mg Co(NO3)2 * 6 H2O 25 mg KI 100 mg MnSO4 * H2O 1540 mg NiCl 1 mg

For use with seeds from green capsules as well as dry seed. Grow seedlings in dark. Refrigerate plantlets for 10 weeks at 5 C while in flask before transfer to soil. [From Anders, via R.J.A.H. Meijer, pers. communication]

A variation, as sent by the author himself:

"About my recipie, you are welcome to give to it to whoever wants to try it. Until now I have had good results with the genera Dactylorhiza, Orchis, Ophrys, Plathantera, Aceras, Serapias, Barlia and Pleione. I use the same medium for germination and replating.

 The medium contains per litre:

MgSO₄ 7H₂0 150 mg

CaCl₂ anhydr. 90 mg

KH₂PO₄ anhydr 150 mg

FeSO₄ 7H₂O 18 mg

Peptone 4 g

Agar 6-8 g

Inositol 100 mg

Pine apple juice 25 ml

Sucrose 14 g

Harvais micronutrients 1 ml

 Harvais micronutrients contains pr litre (1000x concentrated)

H₃BO₃ 500 mg

CuSO₄ 5H₂O 25 mg

ZnSO₄ 7H₂O 500 mg

Na₂MoO₄ 2H₂O 20 mg

Co(NO₃)₂ 6H₂O 25 mg

KI 100 mg

MnSO₄ 1H₂O 1540 mg

NiCl 1 mg

Adjust the pH to 5.7. The peptone should preferably have a low salt (ash) content.

I germinate most species in tightly sealed flasks and transfer the small protocorms to Erlenmeyer flasks capped with aluminum foil, which allow for some air-axchange
